Multi-Well Plates A commonly used approach to study protein expression from cell lines in multi-well plates is dot blotting: samples are transferred from a multi-well plate to amembrane using a vacuum manifold then the blot is analyzed by probing with antibodies. Hair Follicles The hair follicle has been proposed as a surrogate biological system to monitor systemic effects of compounds because it provides a non-invasive system to measure cellular growth control proteins in an individual. The use of hair follicles has the following advantages: samples are easily accessible, follicles contain rapidly dividing cells that are highly susceptible to agents affecting cell growth, several growth control proteins, including growth factor receptors, are important in the growth cycle of hair and are differentially expressed throughout the follicle. Using IHC, hair follicles have been shown to respond to treatment with inhibitors of rTK, a protein kinase involved in cancer.

Tissue Sections and Tissue Microarrays LGS Technology has been applied to analysis of proteins directly in tissue sections. Proteins are identified on each layer with their locations (x-y) conserved from the original sample. The layers are digitized using one of several commercially available laboratory-scanners. Then, using proprietary software developed by 20/20 the layer images are aligned with the image from the original sample on top so that unique protein profiles can be read at any desired location from the tissue sample. Because the layers are vertically aligned with the tissue section, the spatial orientation of the target on the membrane corresponds to its position within the tissue.
